Mechanisms of Cisplatin-Induced Acute Kidney Injury: Pathological Mechanisms, Pharmacological Interventions, and Genetic Mitigations

Administration of the chemotherapeutic agent cisplatin leads to acute kidney injury (AKI). Cisplatin-induced AKI (CIAKI) has a complex pathophysiological map, which been linked cellular uptake and efflux, apoptosis, vascular injury, oxidative endoplasmic reticulum stress, inflammation. Despite research efforts, pharmaceutical interventions, clinical trials spanning over several decades, consistent stable pharmacological treatment option reduce in patients receiving remains unavailable. This predominately incomplete understanding CIAKI pathophysiology molecular mechanisms involved. Herein, we detail extensively known cisplatin-induced nephrotoxicity that manifests variety genetic alteration studies target them.
